The flat oyster Ostrea edulis is the species endemic to European coasts, both Atlantic and Mediterranean. It has been exploited since Roman times, but its aquaculture production decreased from around 20000 tons in the 1950's to 1500 tons today because of two successive diseases due to the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer has been undertaking a breeding program to produce families of oysters tolerant to Bonamia. In this context, a further step would be to identify QTLs for resistance to this parasite. Therefore, as a first step a genetic map is now being built. The establishment of a genetic linkage map will provide a foundation for the mapping of QTLs, with the ultimate objective to implement... |

Background: Although bivalves are among the most-studied marine organisms because of their ecological role and economic importance, very little information is available on the genome sequences of oyster species. This report documents three large-scale cDNA sequencing projects for the Pacific oyster Crassostrea gigas initiated to provide a large number of expressed sequence tags that were subsequently compiled in a publicly accessible database. This resource allowed for the identification of a large number of transcripts and provides valuable information for ongoing investigations of tissue-specific and stimulus-dependant gene expression patterns. These data are crucial for constructing comprehensive DNA microarrays, identifying single nucleotide... |

The most known and certainly the most important contribution of genetics to the oyster indus try is the development of the triploid cupped oyster. Released for the first time in the 1980s, the triploid oyster has triplets of chromosomes instead of pairs. Currently, triploid oysters are obtained by crossing diploid and tetraploid oysters. These oysters are characterized by a near lack of milt in summer, which makes them generally popular with consumers, and by a faster growth, allowing a shorter production cycle. In parallel, the exploitation of natural resistance in the diversity of French oysters has been considered to improve the health of farmed populations which is a major challenge for the sustainability of aquaculture farms. Currently, the... |

The Pacific oyster, Crassostrea gigas, was introduced to Europe for aquaculture purposes, and has had a rapid and unforeseen northward expansion in northern Europe. The recent dramatic increase in number of C. gigas populations along the species' northern distribution limit has questioned the efficiency of Skagerrak as a dispersal barrier for transport and survival of larvae. We investigated the genetic connectivity and possible spreading patterns between Pacific oyster populations on the southern Norwegian coast (4 localities) and Swedish and Danish populations by means of DNA microsatellite analysis of adult oysters, and by simulating larvae drift. In the simulations we used a 3D oceanographic model to explore the influence of recent climate change... |

The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. This latter map now combines the best accuracy and the best estimate of the genome coverage available for an oyster species. Secondly, by comparing the QTLs detected in this study with those previously published for O. edulis in similar experimental conditions, we identified several potential QTLs that were identical between the different families, and also... |

The Pacific oyster, Crassostrea gigas, native to northeast Asia, has been translocated widely for the purpose of aquaculture and is among the most important cultured shellfish. C. gigas has a complex history of introductions in Europe. In France, massive introduction was performed in the early 1970’s, allowing massive spatfall in favourable areas to become the basis of the local oyster industry. In the U.K, hatchery propagation was used to supply local production. Some seed was also exported to Northern Europe were demographically independent populations have been reported in the last decades. In the present study 22 sites were sampled in the British Isles, Denmark, France and Spain in order to assess the genetic diversity and population differentiation... |

Ostreid herpes virus 1 (OsHV-1) infections, notably reported in Europe and the USA, are closely associated with significant mortalities of the Pacific oyster, Crassostrea gigas, especially during its early stages of life. In summer 2006, we monitored mortality by strict daily verification of three full-sib families of oysters reared under common conditions. We quantified OsHV-1 using real-time PCR in dead and living individuals during and after a mortality event. Mortality events were severe and brief, but significantly different between tested families (cumulative mortality ranging from 1.2 to 49%). Real-time PCR assays revealed different viral DNA loads in dead individuals from different families (P < 0.001). Moreover, the mean level of infection... |

Summer mortalities of the Pacific cupped oyster Crassostrea gigas have been described in the literature for decades. One solution to overcome this problem would be to select animals more tolerant to summer mortalities, as this trait was shown to be highly heritable (Degrémont, 2003). Such selective breeding would strongly benefit from the identification of markers associated to the resistance to allow the development of markers assisted selection. Recently, a set of a new kinds of markers (SNPs) have been developed in the Pacific oyster. These markers were identified and characterized in several genes involved in the immune response and energetic pathways (Sauvage, 2007). In this context, the mapping of SNPs, of 16 new EST-SSR based markers and of... |

Exon Primed Intron Crossing (EPIC) markers providemolecular tools that are susceptible to be variable within specieswhile remaining amplifiable by PCR using potentially universal primers. In this studywe tested the possibility of obtaining PCR products from 50 EPIC markers on 23 species belonging to seven different phyla (Porifera, Cnidaria, Arthropoda, Nematoda, Mollusca, Annelida, Echinodermata) using 70 new primer pairs. A previous study had identified and tested those loci in a dozen species, including another phylum, Urochordata (Chenuil et al., 2010). Results were contrasted among species. The best results were achieved with the oyster (Mollusca) where 28 loci provided amplicons susceptible to contain an intron according to their size. This was... |

Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the... |
